murine MAb against Streptokokken-Proteinase, clone HD-SP 4.1
Description
Streptococcal proteinase is a sulfhydryl enzyme elaborated by group A streptococci. It is excreted into the medium as a zymogen (44 kDa). The inactive enzyme (32 kDa) formed by proteolysis of the zymogen requires activation by exposure to reducing agents.
The reduced, active enzyme can be readily inactivated by reagents known to react with sulfhydryl groups.
Preparation
The monoclonal antibodies were generated against purified Streptococcal proteinase. The antibodies have been purified from cell culture supernatant using Protein G affinity chromatography.
Presentation
Screw capped vial containing 1 mg of purified antibody in PBS pH 7.4. The IgG concentration is given on the vial label. Spin the vial briefly before opening.
Storage and Stability
Store the antibody at 2°-8°C. For long-term storage the antibody should be aliquoted and stored at –20°C or colder. It is recommended to avoid freeze-thaw cycles.
Known applications
Western blot
The antibodies are suitable for western blot analysis. They show different reactivity towards the zymogen (Z) or the active proteinase (ST). The clones HD-SP 3.1, 4.1 and 5.1 react solely with the zymogen. Apparently, they recognize an epitope on the 13K-fragment, which is cleaved of during proteolytic activation.
