anti-human CD23, Clone LT-4F1

Description
CD23 (Fc epsilon RII), the low affinity IgE receptor, is a 45 kDa type II membrane glycoprotein expressed more or less on eosinophils, follicular dendritic cells, Langerhans cells, mature B cells (mainly upon activation), EBV­transformed lymphoblasts, monocytes, and subpopulation of platelets. A soluble form of 37 kDa and other its fragments were also described. CD23 mediates IgE-­dependent cytotoxicity by eosinophils and macrophages, and downregulates IgE secretion in response to high levels of IgE, involving release of proinflammatory cytokines.

Properties
The monoclonal antibody ADG5015/L (clone LT­-4F1) is a murine monoclonal antibody, subclass IgG1. The antibody has been purified form ascites by DEAE chromatography, Purity > 95% (by SDS-­PAGE).
The antibody recognizes an epitope located in the stalk region of human low affinity IgE receptor (CD23) between the 37 and 25 kDa cleavage sites.

Presentation
Vial containing 100 µg /100 µl (ADG5015) or 300 µg/ 300 µl (ADG5015L) of purified antibody in PBS containing 0.09 % sodium azide (pH 7.2). The IgG concentration is 1 mg/ml. Spin the vial briefly before opening.

Storage and Stability
Store at 4 °C. For long­term storage aliquot and store at ­-20°. It is recommended to avoid freeze-thaw cycles. The reagent is stable until the expiry date stated on the vial label.

    CD23 is known as the low-affinity IgE receptor (Fc epsilon RII). Binding of IgE to its receptors exerts cellular signaling. Platelets express both, the low-affinity IgE receptor (Fc epsilon RII/CD23), as well as the high-affinity receptor for IgE (Fc epsilon RI). By flow cytometric analysis the authors found that a low proportion of platelets co-expresses Fc epsilon RI and Fc epsilon RII/CD23. Functional studies revealed that anti-Fc epsilon RII/CD23 antibodies significantly inhibited Fc epsilon RI-mediated functions in platelets, indicating down-regulatory effects of receptor-ligand interaction of Fc epsilon RII/CD23.

    The low-affinity IgE receptor (CD23) is expressed by platelets. Thus thrombocytes  may contribute to long-lasting IgE sensitivity by serving as a storage pool for IgE. The authors provide evidence to conclude “that platelets of atopic individuals may contribute to allergic inflammation by serving as a storage pool for IgE and by their increased capacity to liberate further mediators of allergy in response to IgE stimulation.”
