murine MAb against human VAT1, clone HD-pKe 169 3.1

Description
Human Vesicle amine transport protein 1 homolog (VAT-1) is a homologue of the synaptic vesicle membrane protein of Torpedo californica. Its coding gene is located near the BRCA1 locus and thus hVAT-1 may be linked to an inherited predisposition to breast and ovary cancer.

VAT-1-expression was observed in basal keratinocytes of lesional epidermis of bullous pemphigoid patients but not in normal epidermis or in lesional epidermis of pemphigus vulgaris patients. Moreover, hVAT-1 expression in HaCaT cells was found to be calcium-dependent. Normal and malignant mammary and ovary epithelium were found to be hVAT-1-negative.

VAT-1 exerts a specific function in keratinocyte physiology, in particular in calcium-regulated processes, with no evident deregulation in malignancies of the breast and ovary.

Preparation
The monoclonal antibody ADG0011 (clone HD-pKe 169 3.1) is a murine monoclonal antibody, subclass IgG1 recognizing the human VAT1 protein. Mice were immunized with recombinant VAT1. The antibody has been purified from cell culture supernatant using Protein G affinity chromato-graphy.

Presentation
Screw capped vial containing 250 µg of purified antibody in PBS pH 7.4 + 0.01% ProClin300. The IgG concentration is given on the vial label. Spin the vial briefly before opening.

Storage and Stability
Store the antibody at 2°-8°C. For long-term storage the antibody should be aliquoted and stored at –20°C or colder. It is recommended to avoid freeze-thaw cycles.
