murine MAb against uPA B-chain, affinity purified

Description
The monoclonal antibody ADG3689 (clone HD-UK1, subclass IgG1) is a murine monoclonal antibody recognizing human urokinase type plasminogen activator (uPA). The antibody has been purified from cell culture supernatant using Protein G affinity chromatography.

Properties
ADG3689 is directed against a B-chain epitope of human urokinase, near the catalytic site. The antibody reacts with free, and receptor bound, single and two chain (HMW) urokinase, uPA/PAI-1 and, uPA/PAI-2 complexes and the B-chain (33 kDa) fragment.

Presentation
Screw capped vial containing 250 µg of purified antibody in PBS pH 7.4, 0.01 % ProClin300. The IgG concentration is 1 mg/ml. Spin the vial briefly before opening.

Storage and Stability
Store the antibody at 2°-8°C. For long-term storage the antibody should be aliquoted and stored at
-20°C or colder. It is recommended to avoid freeze-thaw cycles.

Applications
A. ELISA
The antibody can be used as detecting antibody in ELISAs. An antibody concentration of 1µg/ml is recommended (1, 2).
B. Immunohistochemistry
The antibody is suitable for staining frozen acetone fixed skin sections (3), and formalin fixed, paraffin embedded sections of cancerous and normal human breast and colon tissue where urokinase is present. Working dilutions range from 1:100 (4) to 1:25 (6).
C. Flow cytometry
The antibody can be used for flow cytometric analysis. An antibody concentration of 10 µg/ml is recommended (7).
D. Western Blot Analysis
The antibody is suitable for Western blot analysis, detecting pro-uPA, HMW-uPA and LMW-uPA after electrophoresis under non-reduced conditions and B-chain fragments under reducing conditions at a working concentration of 1 μg/mL (8).
E. Inhibition
The antibody was shown to inhibit plasminogen activation on the surface of melanoma cell lines (9).
