murine MAb anti-human TFPI Kunitz domain 1

Description
The monoclonal antibody ADG4903 (clone 2H8) is a murine monoclonal antibody, subclass IgG1 recognizing human Tissue factor pathway inhibitor (TFPI). Mice were immunized with purified native human tissue factor pathway inhibitor (TFPI, 38 kDa) from cultured HEP G-2 cells.(1) The epitope specificity of this antibody is aa 22-87, within Kunitz domain I of human TFPI which is involved in the TF/fVIIa interaction. This antibody neutralizes human TFPI, see Applications, below.

Presentation
Screw capped vial containing 100 µg of purified antibody in PBS pH 7.4, 0.01 % ProClin. The IgG concentration is 1 mg/ml. Spin the vial briefly before opening.

Storage and Stability
Store the antibody at 2°-8°C. For long-term storage the antibody should be aliquoted and stored at -20°C or colder. It is recommended to avoid freeze-thaw cycles.

Applications
Western Blot Analysis: at 5 μg/mL the antibody will detect 50 ng/mL of human TFPI under nonreducing conditions only. (2)
Immunohistochemistry: the antibody is suitable for staining of human TFPI in formalin-fixed tissue sections. (3, 4)
Flow cytometry: the antibody was used for staining of cells for flow cytometric analysis. (4)
Neutralization of human TFPI inhibition (complexed with human FXa, product no. 526): at 5 μg/mL the antibody neutralizes TFPI at 5 ng/mL
The antibody prevents the inhibition of the TF-FVIIa function. (5)
